Speak to Fresh in Karakura Town by the HospitalObtain the quest to defeat a ShinigamiDefeat the Shinigami and pick up the keyWalk into the Hospital and make your way to Room 9Use the key to open the door to Room 9Pick up the Quincy Book inside the roomGo outside to meet DoudaAfter being teleported to Hueco Mundo, talk to Tyrone to receive a quest (collect a Hollow Mask!)Defeat a Hollow, pick up its mask, and take it to TyroneYou’ll be teleported to Yhwach who will give you the Quincy blood (this has an 80% success rare – failure means you’ll have to repeat the process)
